For its price I think the Rome Hop-on Hop-off Double Decker Bus Tour is the best little tour of Rome you can get. Being on the top deck means you get a great perspective of some of the greatest monuments in the city. The audio commentary–despite not being recorded and not from a live guide–comes on at the right moment and is clear and very informative.

Some parts of the route are really very special, particularly heading down the Via della Concilianze with St. Peter’s Basilica looming up ahead of you. The other great part is when you leave Termini and pass St. Maria Maggiore (one of the more important churches of Rome), go by the Quirinale (a majestic Roman palace with impressive obelisk and statues in the square), then down the Via Nazionale and into chaotic Piazza Venezia with the huge “wedding cake” looming in front of you. And if that wasn’t enough, you have a great long build up to the Colosseum by going down the Via dei Fiori Imperiale.

The fact that you can get on or off at any of these major sites without incurring any additional fees really makes this a very informative and scenic way of getting from one site to another or just simply going for a bit of a scenic wander. Tickets begin in the 24-hour validity category upwards.
